Thin-layer chromatographic determination of L-ascorbic, L-dehydroascorbic nd 2.3-diketo-L-gulonic acids in animal tissues, blood and urine.

چکیده
A new method for the determination of ascorbic, dehydroascorbic and 2.3-diketogulonic acids in animal tissues, blood and urine is described. The material for analysis is homogenized in the presence of stannous chloride in trichloracetic acid and the extract is divided into three aliquots: in one part the ascorbic acid is converted into dehydroascorbic acid by oxidation with bromine water, in the second part the original content of dehydroascorbic and diketogulonic acids is stabilized by the addition of thiourea and in the third part dehydroascorbic acid is reduced to ascorbic acid by thioglycolic acid. Reaction with 2.4-dinitrophenylhydrazine produces a mixture of osazones, from which the bis-hydrazone of diketogulonic acid is isolated by thin-layer chromatography on Silicagel (solvent, ethyl aeetate-chloroform. acetic acid 50:50:5 v/v). The amount of bis-hydrazone is determined photometrically at 502 nm after dissolution in ethyl-acetate-toluene (1:3 v/v). The content of ascorbic, dehydroascorbic and diketogulonic acids is calculated by comparison of the results from the three aliquots of the extract. The described method is more specific and more sensitive than the former colorimetric methods and is suitable for the isolation and evaluation of small amounts of ascorbic, dehydroascorbic and diketogulonic acids labelled with carbon C.

متن کاملFormation of L-lyxonic and L-xylonic acids from L-ascorbic acid in rat kidney.
The enzymatic decarboxylation of n-ascorbic acid by rat kidney homogenates has been reported in an earlier study (1). It was shown that factors present in both the soluble and the particulate fractions were required for maximal activity. Evidence was presented suggesting that dehydro-L-ascorbic acid and 2,3-diketo-n-gulonic acid were intermediates in the decarboxylation of the vitamin. The reac...

Elson (1943) found that urease is inhibited by small amounts of ascorbic acid and that this inhibition is eliminated in the presence of cysteine. Quastel (1943) suggested that the inhibition was due to a reaction between dehydroascorbic acid and an -SH group of the enzyme, and that the protective effect of thiol compoundcls was due to the fact that they reduced dehydroascorbic acid. متن کاملThe Oxidative Degradation of L-Ascorbic Acid via an a-Ketoaldehyde *
Dehydro-L-ascorbic acid, the oxidation product of L-ascorbic acid, is unstable in aqueous solution and decomposed by a hydrolytic disruption of its ring structure to 2,3-diketo-L-gulonic acid. It is shown by means of UV, 'H-, and 13C-NMR spectroscopy that after decarboxylation 3,4,5-trihydroxy-2-keto-L-valeraldehyde, an a-ketoaldehyde, is formed. This substance is oxidized further to L-erythroa...
